3D-Aldehyde Microplates

for covalent binding of amine-containing biomolecules

Aldehyde groups react immediately with the amine groups of biomolecules to form an imine (Schiff base). To increase the bond strength it is possible to reduce the imines with e.g. Sodium Borhydride (NaBH4) or Tris(2-carboxyethyl)phosphine (TCEP) to form stable secondary amines.

Upon completion the coupling reaction other non-reacted aldehydes must be blocked with small molecules that penetrate the 3D-Matrix and effectively quench all remaining reactive groups.
